Содержание циркулирующих CD4+FOXP3+ Т-клеток было исследовано у 59 больных ММ, которым проводили ВХТ с ауто-ТГСК. Количество CD4+FOXP3+ Т-клеток оценивали методом проточной цитометрии до ВХТ с ауто-ТГСК на момент выхода из лейкопении через 6 и 12 мес. Количество CD4+FOXP3+ Т-клеток полностью восстанавливалось и значимо превышало исходную концентрацию ко дню выхода из лейкопении. В последующем (в течение года) их относительное содержание постепенно снижалось до нормы. Анализ взаимосвязи CD4+FOXP3+ Т-клеток с исходами ауто-ТГСК показал, что больные с рецидивом ММ в течение первых 12 мес после ауто-ТГСК различались значимо более высоким относительным содержанием CD4+FOXP3+ Т-клеток на момент выхода из лейкопении. Проведение анализа характеристических кривых (receiver operating characteristic – ROC) выявило высокую прогностическую значимость оценки CD4+FOXP3+ Т-клеток как предиктора раннего развития рецидива заболевания после ВХТ с ауто-ТГСК. Таким образом, количество циркулирующих CD4+FOXP3+ Т-клеток после ауто-ТГСК восстанавливается быстро и полностью. Развитие раннего рецидива заболевания после ВХТ с ауто-ТГСК ассоциировано с более высоким содержанием CD4+FOXP3+ Т-клеток ко времени выхода из лейкопении. Повышенное содержание этих клеток позволяет прогнозировать развитие рецидива ММ в раннем посттрансплантационном периоде.</p></abstract><trans-abstract xml:lang="en"><p>We have investigated the dynamics of post-transplant recovery of CD4+FOXP3+ T cells in patients with multiple myeloma (MM). The relationship between CD4+FOXP3+ T cell counts and clinical outcomes of MM patients after high-dose chemotherapy and autologous hematopoietic stem cell transplantation (AHSCT) also has been evaluated. Fifty-nine patients were included in the study. The content of circulating CD4+FOXP3+ T cells has been assessed with the use of flow cytometry before AHSCT, at the day of engraftment, and 6 and 12 months later. After AHSCT the relative count of CD4+FOXP3+T cells in MM patients restored rapidly, it became higher than initial level at the day of engraftment and then subsequently decreased for a 1 year to values in healthy donors. A relative count of CD4+FOXP3+T cellsat the time of engraftment increased in patients with relapsed disease during 12 months after AHSCT.
