Цикл производства Т-лимфоцитов с химерным антигенным рецептором (chimeric antigen receptor, СAR) включает в себя несколько последовательных стадий, параметры которых могут влиять на эффективность трансгенных клеток: получение клеточного материала больного, выделение целевой популяции Т-лимфоцитов, активация, трансдукция клеток вирусным вектором, несущим CAR конструкт, и экспансия полученных CAR T-клеток с дальнейшим введением больному.</p></sec><sec><title>Цель</title><p>Цель: рассмотреть влияние каждого из этапов производства CAR T-клеток на показатели эффективности клеточного продукта в in vitro и in vivo экспериментах, а также при клиническом применении.</p></sec><sec><title>Основные сведения</title><p>Основные сведения. Проведен анализ особенностей производства различных CAR T-клеточных препаратов. Обсуждено, как характеристики производства препарата и структуры химерного антигенного рецептора влияют на противоопухолевую эффективность и профиль безопасности клеточного продукта. Получение CAR T-клеточного продукта — многофакторный процесс, который требует оптимизации параметров и обязан учитывать особенности исходного сырья.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. The production cycle of CAR T-cell product includes several sequential stages, each of which may inﬂ uence the efﬁciency of transgenic cells: obtaining the patient’s cellular material, isolating the target T-lymphocyte population, activation, transduction of the cells with a viral vector carrying the CAR construct, and expansion of the obtained CAR T-cells with further administration to the patient.</p></sec><sec><title>Aim</title><p>Aim: to review the impact of each of the production steps on the cell product performance in both in vitro and in vivo experiments, as well as clinical applications.</p></sec><sec><title>Main ﬁndings</title><p>Main ﬁndings. The manufacturing characteristics of various CAR T-cell products were analyzed in this review, followed by a discussion on how different manufacturing characteristics and chimeric antigen receptor structures affect the antitumor efﬁcacy and safety proﬁ le of the cell product.
