Высокоиммуногенный антиген D — один из наиболее важных после групповых антигенов A и B. Многочисленные аллели гена RHD приводят кпоявлению новых фенотипов антигена D. Фенотип Del характеризуется сверхслабой экспрессией антигена D.</p></sec><sec><title>Цель</title><p>Цель: представить результаты типирования антигена D у донора крови с фенотипом Del.</p></sec><sec><title>Материалы иметоды</title><p>Материалы иметоды. Для серологического типирования крови донора использовали технологию микроколоночной агглютинации и твердофазную микропланшетную технологию «Capture». Первичное генотипирование донора выполняли методом аллель-специфичной полимеразной цепной реакции (АСП-ПЦР). Был проведен поиск нуклеотидных замен методом секвенирования по Сэнгеру продуктов ПЦР экзонов гена RHD.</p></sec><sec><title>Результаты</title><p>Результаты. При проведении серологического типирования с анти-D-антителами IgM и IgG с использованием твердофазной микропланшетной технологии был получен положительный результат. При определении парциальных вариантов антигена D однозначно интерпретировать полученные данные не получилось. Для идентификации вариантов антигена weak D потребовалось проведение молекулярно-генетического анализа. По результатам первичного генотипирования методом АСП-ПЦР было сделано предположение о делеции 9-го экзона гена RHD либо нуклеотидной замене в 9-м экзоне. По результатам секвенирования по Сэнгеру в экзоне 9 был обнаружен вариант нуклеотидной последовательности ДНК с. 1203T&gt;A, приводящий к образованию стоп-кодона Tyr401Ter (rs759513820). Данный генетический вариант соответствует аллелю RHD*01EL.17. Антиген, кодируемый этим аллелем, относится к типу Del.</p></sec><sec><title>Заключение</title><p>Заключение. Необходимо применять иммуногематологические методы в сочетании с молекулярно-генетическими методами для типирования антигена D у доноров крови с фенотипом Del с целью предотвращения аллоиммунизации антигеном D.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. The highly immunogenic D antigen is one of the most important antigens after the group antigens A and B. Numerous alleles of the RHD gene lead to the emergence of new D antigen phenotypes. The Del phenotype is characterized by an extremely weak expression of the D antigen.</p></sec><sec><title>Aim</title><p>Aim: to present the results of D antigen typing in a blood donor with the Del phenotype.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Two technologies were used for serologic typing of donor blood: microcolumn agglutination technology and solid phase microplate Capture technology. Primary genotyping of the donor was performed by allele-specific polymerase chain reaction (ASP-PCR). Nucleotide substitutions were searched for by Sanger sequencing of PCR products of exons of the RHD gene.</p></sec><sec><title>Results</title><p>Results. When serological typing with IgM and IgG anti-D antibodies using solid-phase microplate technology was performed, a positive result was obtained. When determining partial variants of the D antigen, it was not possible to unambiguously interpret the data obtained. Molecular genetic analysis was required to identify variants of the weak D antigen. The results of primary genotyping by ASP-PCR suggested deletion of exon 9 of the RHD gene or nucleotide substitution in exon 9. According to the results of Sanger sequencing, a variant of the DNA nucleotide sequence c.1203T&gt;A was detected in exon 9, resulting in the formation of the stop codon Tyr401Ter (rs759513820). This genetic variant corresponds to the RHD*01EL.17 allele. The antigen encoded by this allele is of the Del type.</p></sec><sec><title>Conclusion</title><p>Conclusion.
